Standard Vitamin D gegen GBM Zellen

Vor einiger Zeit auf dem ESMO Patientenkongress gabs auch einige Verlage, die ausstellten.

Hab da einige interessante Bücher zur komplementären Therapie gekauft.
Ein ganzes Buch fand ich zu Vitamin D.
Mit einem Kapitel zu Hirntumoren.
Vitamin D kann die entarteten Zellen zum Selbstmord bringen..

Ich habe Vitamin D (Tropfen) schon immer genommen. Es gibt viel Studien dazu in der pubmed:

Induction of apoptosis by vitamin D metabolites and analogs in a glioma cell line.

Gliomas are the most common malignant tumors in brain. Recent studies demonstrate the capacity of 1alpha,25(OH)2D3 to specifically induce cell death (apoptosis) in model glioma cell lines and in primary cultures from tumor tissue, but not in primary astrocytes. In spite of this promising activity, a broad therapeutic application of vitamin D metabolites and analogs is still restricted because of their poor bioavailability and their hypercalcemic actions. Compared to 1alpha,25(OH)2D3, its natural 3alpha-epimer exhibits far higher metabolic stability and a reduced calcemic effect. Focusing on a possible therapeutic advantage of the 3alpha-conformation, we have examined the apoptotic potential of a representative set of vitamin D analogs, each of them in the 3alpha- and 3beta-conformation, and of natural vitamin D metabolites in the rat C6 glioma cell line. Exposure of these cells to the synthetic analogs resulted in all cases in a pronounced reduction of cell density (tested by incorporation of neutral red) and induction of apoptosis, monitored by staining nuclei with Hoechst 33258 dye and by following DNA fragmentation by capillary electrophoresis. The 3alpha-epimers showed equivalent or even higher activity on C6 cells than their respective 3beta forms. For their potent effects on growth and apoptosis of tumor cells and their high metabolic stability combined with a low calcemic potential, we speculate that these 3a-epimers could provide advantages for a prospective treatment of glioma.

Hallo Manfred,

ja in dieser Richtung wird viel geforscht und es gibt in Zukunft bestimmt auch einige Ansätze.

Nur ich warne eigendlich davor solche "Eigentherapien" zu machen, denn aus diesem Artikel geht hervor das es bis jetzt nur Versuche an Ratten gegeben hat.
Das heißt für uns es gibt keinerlei Erfahrungswerte wie hoch das Vit. D genommen werden muß.
Leider ist es so, das zwischen den Tierversuchen und dem Menschen doch sehr viele Unterschiede liegen... was beim Tier wirkt, wirkt noch lange nciht beim Menschen...

Das Vit. muß in hohen Dosen genommen werden, nur wann beginnt der lebensbedrohliche Bereich?


Auch ist inzwischen klar, das hoch dosierte Vitamine nicht bei Chemos eingenommen werden sollen, denn Zellen die mit Vitaminen vollgestopft sind lassen dem Zytostatika nur wenig Möglichkeit sich an die Zelle zu binden.

PURPOSE: Classical and new therapies in anaplastic astrocytomas and glioblastomas do not yield sufficient results. Agents able to redifferentiate neoplastic cells in vitro are known. We proposed alfacalcidol, a vitamin D analog able to bind to nuclear receptors regulating mitotic activity, in the treatment of malignant gliomas.

PATIENTS AND METHODS: Patients with glioblastomas and anaplastic astrocytomas were enrolled in a phase II trial involving surgery or biopsy, radiotherapy (64 Gy), chemotherapy with VM26-CCNU or fotemustine, and alfacalcidol at the daily dose of 0.04 microg/kg. MRI took place every 6 months. RESULTS: Eleven patients were included and completed the study. The series involved 10 glioblastomas and 1 anaplastic astrocytoma. Three patients out of 11 patients (27%), 2 glioblastomas and 1 astrocytoma grade III, exhibited a particular response, consisting in the progressive regression of the radiological lesion, with a decrease of the gadolinium-enhanced area. Simultaneously, the patients showed a complete clinical remission, observed respectively for 7, 5 and 4 years. In the series of 10 patients with glioblastomas, 2 cases showed this response; after 4 years, 2 of 10 patients with glioblastomas (20%) were alive; the median survival time is 21 months. Normal or subnormal calcemia was observed, at the dose proposed, so that no interruption of the drug was necessary.

CONCLUSIONS: Alfacalcidol, an in vitro agent of redifferentiation, is safe and seems able to induce in some patients, in synergy with classical surgery-radiotherapy-chemotherapy treatments, a particular progressive and durable regression of the tumor. The responders might represent about 20% of malignant gliomas.

Hallo Manfred,

das hört sich nciht schlecht an.

Nur bei 11 Patienten in einer Studie kann man natürlich nicht viel sagen..
Dann frage ich mich, es war eine Phase 2 Studie, die 2001 durch geführt wurde.

Warum hat man nicht auch eine Phase drei durchgeführt...??

Eigendlich Schade...

weiß nicht, könnte daran liegen, dass es sich kommerziell nicht gut nutzen läßt.
Wär ja viel zu simpel ausserdem, bloss ein Vitamin.

Mir hat man übrigens nach meiner OP im März Roaccutan vorgeschlagen.
Zum Glück hab ich s nicht gemacht.
Hätte ich gleich in Hochdosis Vitamin A nehmen können. Roaccutan ist ein iosmerisiertes Vitamin A, und ist dadurch éxtrem teuer. Eigentlich ein schweres Aknemittel, das man auch mal bei Hirntumor ausprobieren wollte.

Ich hab dann später erfahren, dass es ausser schweren Nebenwirkungen (aufgebogene schmerzhafte Finger und Zehennägel, Wundwerden der Haut und Lippen etc.) nichts gebracht hat.
Manche Tumore sind dann sogar noch schneller gewachsen.
